The co-culture system of primary rat hepatocytes (PRH) with Madin-Darby canine kidney cells (MDCK) is a useful model system for the toxicological evaluation of drug-induced liver injury. However, the suspension culture of PRH by itself is limited, because MDCKs are anchorage dependent and remain attached to the bottom of the culture vessel. This study aimed to develop a hepatocyte-based microcarrier for the co-culture of PRH and MDCK. Hepatocyte spheroids were fabricated using a gas-liquid-interface microfluidic chip and attached to a polyacrylamide-based microcarrier (microcarrier-HCs). The microcarrier-HCs were successfully immobilized in the culture medium using a poly-L-lysine (PLL) coating. The immobilized microcarrier-HCs were then cultured with MDCK cells. The attachment of MDCK cells on microcarrier-HCs was suppressed compared to HCs because the attachment of MDCKs on microcarrier-HCs was initiated after cell detachment from microcarrier-HCs. The attachment of MDCK cells on microcarrier-HCs was slightly suppressed by the presence of hepatocytes. After 7 days of co-culture, the incorporation of albumin into the media, the expression of CYP3A1 and CYP3A2, and the expression of the CYP2E1-dependent activity of 7-ethoxyresorufin-O-deethylase (EROD) were increased in MDCK cells. The results suggest that microcarrier-HCs successfully support the co-culture of PRH and MDCK cells in a Petri dish and that they provide a useful model for the assessment of hepatocyte toxicity.
